As another poster said, I'd recommend a clean install and re-build if at all possible in either case. You'll be floored by how much faster the system I want to replace my single hard drive with a larger one. I don't want to keep the old drive. What is the easiest way to copy everything over to the new drive?

If you put it on the same ribbon as a CD-ROM (or CD-R) leave the hard drive alone and make the CD device slave. It is easier to install 3.5'' drives in 3.5'' bay, and likewise with 5.25'', but you could use a mounting thing to put a 3.5'' in a 5.25'' bay, though I don't recommend it. Screw it in.

Thanks for the help, probaby something small I am missing - I installed VistaBootPro and run it from my Vista install, should I install it and run it from the XP install instead? I installed it, by booting from the Vista DVD, selecting the new hard drive, formatted it from there, and installed Vista onto it.
